Mesenchymal stem cells desensitize castration-resistant prostate cancer to docetaxel chemotherapy via inducing TGF-β1-mediated cell autophagy

Abstract Background Mesenchymal stem cells (MSCs) have been proved to drive castration resistant prostate cancer (CRPC). In this study, we aim investigate the contribution of MSCs development docetaxel resistance in CRPC and its potential mechanisms. Methods The effect on was determined using vivo vitro approaches. CCK8 PI/Annexin V-FITC assay were used examined cell viability apoptosis. concentration transforming growth factor-β1 measured by enzyme-linked immunosorbent small interfering RNA for functional analyses. Results significantly reduced sensitivity docetaxel-induced proliferation inhibition apoptosis promotion vitro. cocultured with under administration an increased autophagy activation, while inhibitor could effectively reversed MSCs-induced docetaxel. Additionally, increase depends secreting TGF-β1 secretion consequently Conclusions These results suggest that administrated may elicit increase, which desensitizes chemotherapy accelerating chemoresistance occurrence via inducing autophagy.
